Ram slots - can they be turned off/on

I have a E-machines t5048 WHICH i PURCHASED  the correct additional ram for my computer and when I installed the new stick, the computer would not boot up.  MY bios is up to date.  I wonder if the slot us turned off and needs to be turned on..  Tried both stick in slot 0 and both worked fine.

I don't believe this issue is related to a slot being turned off.  Have you tried a single module alone in the second slot?  The system should boot no matter what slot you put the memory in.  When installing, make sure to press with a good 20 to 30 pounds of pressure so the clips close completely on their own.
